application
4, 6-dihydroxy-2-methylpyrimidine can be used as an intermediate in organic synthesis. The compound is used to produce explosives -1, 1-diamino-2, 2-dinitroethylene (DADNE,FOX7), and its application in the synthesis of biologically active substances can also be found.
Preparation
4, 6-dihydroxy-2-methylpyrimidine is prepared as follows: in a reaction flask equipped with a mechanical stirrer, reflux condenser, thermometer and dropping funnel, put 200ml of isopropanol, pour 27g(0.5mol) of sodium methoxide, and heat at 40-50°C until completely dissolved, then add 20ml (0.125 mol) of malonate, mix for 10-15 minutes, then 22 grams (0.373 moles) of acetamide were added in batches under vigorous stirring. The reaction material is quickly heated to boiling and boiled for 4 hours, then the solvent is distilled, and 100ml of water is added to the residue at a temperature of 50-60°C, and the precipitate is completely dissolved. The resulting salt solution was acidified with glacial acetic acid at 50 ÷ 60°C to a pH of 5-6, slowly cooled and settled at a temperature of 18-25°C for 10 hours. Filter out the precipitated product and wash with water (3 × 30ml) and alcohol (2 × 20ml). 9.9g (62.9% of the theoretical value) of 4, 6-dihydroxy-2-methylpyrimidine with a basic substance content of 95% was obtained.
