Abstract:
Objective: to determine the content of Daunorubicin Hydrochloride in liposomes. Method: using C_(18) column as analytical column, acetonitrile -0.02 mol · L ~(-1) sodium dihydrogen phosphate solution-triethylamine (34:66:0.3, adjusted to pH 4.0 with phosphoric acid) the flow rate was 1.0 mL · min ~(-1), and the detection wavelength was 233nm. The sample volume was 50 μL. Results: in the range of 2.51~25.10 μg · mL ~(-1), the peak area (A) and concentration (C) of Daunorubicin Hydrochloride there is a good linear relationship (r = 0.9993). The detection limit was 40ng · mL ~(-1). The average recoveries of low, medium and high concentrations were 100.8%,103.4% and 102.2%(n = 5) with RSD of 0.49%,0.42% and 1.9%, respectively. Intra-day, inter-day RSD(n = 5) were 1.1%-1.6% and 0.60%-1.5%, respectively. Conclusion: The method is rapid, simple and specific for the determination of Daunorubicin Hydrochloride in liposomes.Key words:

Abstract:
The Electrochemical Behavior of Daunorubicin Hydrochloride and its interaction with bovine serum albumin (BSA) were studied by linear sweep voltammetry and cyclic voltammetry. The results showed that Daunorubicin Hydrochloride had a sensitive reduction peak with a peak potential of Britton-Robinson in Ep-0.64V buffer solution at PH 6.50 (. The reduction peak current of Daunorubicin Hydrochloride decreased after the buffer solution was added to BSA. Based on this, an electrochemical method for the determination of BSA was established. Under the optimized conditions, the peak current was linear with the concentration of BSA in the range of 1.0 × 10-8~1.0 × 10-4mol · L-1(r = 0.9965), and the detection limit was 5.0 × 10-9mol · L-1. The binding ratio and binding constant of Daunorubicin Hydrochloride to BSA were also determined.
