preparation | a volume of 3, 5-difluoro anisole was dissolved in of glacial acetic acid, 7.6ml of acetic anhydride, A mixture of 3.2ml fuming nitric acid and 3.2ml glacial acetic acid was slowly added dropwise in a water bath at 50-60 ℃. After completion of the dropwise addition, the mixture was heated in a 60 ° C. Water bath and stirred for 1H. The reaction mixture was then poured into 400ml of water, stirred and allowed to stand overnight. The yellow semi-solidified oily liquid in the mixture was filtered off, recrystallized from 95% ethanol and left in a refrigerator overnight to obtain 5.10g of pale yellow solid III, yield 47.7%, melting point 137.2 °c. Weigh 1.50g (about 7.9mmol) of III and dissolve in of methanol, then weigh 6.8g of sncl2 and dissolve in 75ml of concentrated hydrochloric acid, mix the above two solutions, and stir at room temperature for 12 hours. A white precipitate formed in the solution was filtered and the solid was washed with a small amount of concentrated hydrochloric acid and dried to give 1.03g of 2, 6-difluoro-4-methoxyaniline. |
