xanthine hydride is an important raw material for oligonucleotide synthesis, alias 3-amino -1,2, 4-dithiazole-5-thione, molecular formula C2H2N2S3, yellow crystal, commonly used in organic synthesis, it is one of the commonly used thioreagents in the solid phase synthesis of thioantisense oligonucleotides. Phosphorothioate antisense oligonucleotides are a class of antisense oligonucleotide analogs formed by the substitution of non-bonded oxygen atoms on the phosphate backbone of antisense oligonucleotides with sulfur atoms, it overcomes the property that antisense oligonucleotides are degraded by enzymes in serum, and thus has an important role in the treatment of cancer and the inhibition of gene expression. In the solid-phase synthesis of thiooligonucleotides, once the oligonucleotides on the carrier are coupled to phosphoramidite riboside, the nucleotides on the new coupling need to be thiooxidized, the yield of the thioreaction affects the purity of the product of the thioantisense oligonucleotide, and the incomplete thiogeneration will lead to the introduction of Oxo impurities, therefore, the quality of the thioreagent has an important influence on the synthesis of the thioantisense oligonucleotide and the content of the final product.
