In vitro study | In SIRT2-myc-expressed HeLa cells, AGK2 potently inhibits SIRT2 activity and increases acetylation of tubulin. In primary midbrain medium, AGK2 protects dopaminergic neurons from α-syn-induced toxicity. AGK2 induces cell necrosis and caspase-3-dependent apoptosis in C6 glioma cells. SIRT2 also reduced the viability of merlin-mutant mouse schwann cells (MSCs) without substantially reducing the viability of wild-type MSC. |
In vivo study | In a Drosophila model of Parkinson's disease, AGK2 scavenges α-syn-mediated toxicity and amends polymerization. |
