Production method | Pyridine is oxidized with hydrogen peroxide, phthalic anhydride is added to phthalate, and then decomposed with dilute hydrochloric acid to make N-oxidized pyridine hydrochloride. N-oxypyridine hydrochloride is nitrated to produce N-oxy-4-nitropyridine. N-oxy-4-nitropyridine is reduced by iron powder to obtain 4-aminopyridine. Dissolve 1gN-oxy-4-nitropyridine (melting point 159 ℃) in 30ml of acetic acid, add 2.2g of iron powder, and the reaction starts immediately. After adding, keep the 100% for 1h and cool. After adding about 50ml of water to dilute, neutralize with 30% sodium hydroxide solution and adjust to strong alkaline, extract with ether 3 times, 100ml each time. The ether extract was combined and dried with anhydrous potassium carbonate. After the ether is evaporated, the crude 4-aminopyridine is distilled under reduced pressure. Recrystallization of the crude product from benzene or toluene-petroleum ether mixed solvent yields 0.6g of white needle crystal 4-aminopyridine with 90% yield. |

N-oxo-4-nitropyridine was prepared by nitration of N-oxo-pyridine hydrochloride. 4-nitropyridine N-oxide is reduced by iron powder to give 4-aminopyridine. 1g of n-4-nitropyridine oxide (m. P. 159 ° C.) was dissolved in 30ml of acetic acid, 100% g of iron powder was added, and the reaction was started immediately. After the addition was completed, the mixture was kept for 1H and allowed to cool. After dilution by addition of about 50ml of water, neutralized with 30% sodium hydroxide solution and made highly basic, extracted three times with of ether each time. The ether extracts were combined and dried with anhydrous potassium carbonate. After the ether was distilled off, the crude 4-aminopyridine was distilled off under reduced pressure. The crude product was recrystallized from benzene or a mixed solvent of toluene and petroleum ether to obtain 0.6g of white needle-like crystalline 4-aminopyridine in a yield of 90%.
