Abstract:
Objective: to establish a method for determination of piperaquine phosphate in human plasma by high performance liquid chromatography with ultraviolet detection. Methods: plasma samples were basified with 1 mol · L ~(-1) sodium hydroxide solution, extracted with diethyl ether, absorbed organic phase, dried by N_2 gas flow, and reconstituted with mobile phase. Alltima C_(18) column (4.6 × 150, 5 μm), mobile phase was acetonitrile-0.1%(w/v) trichloroacetic acid-phosphoric acid (25:75:0.035,v/v/v), the flow rate was 0.4 mL · min ~(-1), the column temperature was room temperature, the detection wavelength was 345 nm, and chloroquine phosphate was used as internal standard. Results: piperaquine phosphate, internal standard and endogenous impurities were well separated. The linear range was 0.02~1.0 μg · mL ~(-1)(r = 0.9993), the lower limit of quantitation was 0.01 μg · mL ~(-1), the intra-day and inter-day RSD was less than 12%, the extraction recovery was 69.3% ~ 76.3%, and the method recovery was 93.4% ~ 106.5%. Conclusion: The method is accurate, stable, sensitive and suitable for the Study of pharmacokinetics and bioavailability of piperaquine phosphate.expand

Abstract:
Objective to establish an HPLC method for the determination of related substances in piperaquine phosphate and to investigate the stability of piperaquine phosphate against light. Methods an Agilent C18 column was used, with 0.04mol.L-1 potassium dihydrogen phosphate-acetonitrile-triethylamine (90:10:0.2, adjusted to pH 2.5 with phosphoric acid) as mobile phase, flow rate: 0.5mL.min-1, detection wavelength: 230nm, column temperature: 25 ℃. Results the content of related substances of piperaquine phosphate was 0.21% ~ 0.39%, which was significantly increased after 10 days of strong light irradiation. Conclusion The method is accurate, reliable, simple and rapid, and suitable for the quality control of piperaquine phosphate.Key words:
