5(6)-HEX - Introduction

5(6)-HEX(5(6)-HEX) is a commonly used fluorescent dye with a molecular formula of C21H12Cl6O5S and a relative molecular mass of 534.16g/mol. It is a yellow to orange solid or powdery substance in organic solvents.

5(6)-HEX is widely used as a cell and tissue marker in fluorescence microscopy and flow cytometry. It can be combined with amino compounds, especially the amino groups of proteins, to form stable fluorescent dye-conjugates. Therefore, it is often used in biomedical research and immune cell analysis.

5(6)-HEX can be prepared by two methods. One is the reaction of fluorescein -5-isothiocyanate with diethylamine to form 5(6)-HEX-fluorescein -5-isothiocyanate salt, which is then reacted with an amino compound to form 5(6)-HEX. Another method is to react a fluorescein steric compound with cuprous bromide and then with an alcohol or phenol to give 5(6)-HEX.
