| synthesis method | step 1: treat 5-bromopyridine -2-carboxylic acid (15.0g,74.3% mmol,Alfa) and DIPEA(38.8 mL) with N-methyl O-methyl hydroxylamine hydrochloride (7.24g,74.3 mmol), 222.8 mmol), HOAT(1.01g,7.43 mmol),EDCI(21.4g,111.4 mmol) and DMF(150 mL) solution. Stir the mixture at room temperature for 48 hours, then add water (100 mL). The mixture was concentrated to about 50 mL under high vacuum and diluted with 300 mL of toluene. Separate each layer and use water (100 mL) and 0.1 N HCl(100 mL). The organic layer was dried (MgSO4) and concentrated to provide 67 a1 for use in the next step. 67a 1. Step 2: Dissolve amide 67 a1(919.0g,40.8 mmol) in THF(100 mL) and cool to -78°C. The mixture was treated with MeLi(31.6 mL,44.9 mmol) and stirred for 1 h at this temperature. The reaction was quenched with saturated NH4Cl and the solvent was evaporated. The residue was alkalized with saturated NaHCO3 and extracted with EtOAc(2x).
