Use | Protocatechuic acid is a phenolic acid substance naturally occurring in many vegetables and fruits. It is also one of the effective active ingredients of Chinese medicinal materials such as Acorus calamus and Yizhi Ren. The water-soluble phenolic acid component procatechuic acid (protocatechu icacid,PCA) is widely found in common traditional Chinese medicines such as geranium salvia miltiorrhiza and classic compound preparations such as compound salvia miltiorrhiza injection and Guanxinning injection. It has expectorant and antiasthmatic effects and is clinically used for the treatment of chronic bronchitis. In addition, it also has significant antioxidant, reduce myocardial oxygen consumption, improve myocardial oxygen tolerance, inhibit tumor cell growth and induce HL60 leukemia cells, HepG2 liver cancer cells apoptosis, slow down the heart and other pharmacological effects, and a large number of literatures reported that it has a certain impact on stem cell proliferation. Protocatechuic acid can promote the proliferation of human adipose-derived stem cells, while still maintaining the characteristics of the multi-differentiation potential of mesenchymal stem cells, inhibiting cell apoptosis, improving cell viability, and may have therapeutic effects on oxidative stress-related neurodegenerative diseases. It may also be combined with human adipose-derived stem cell transplantation for the treatment of neurodegenerative diseases. In recent years, studies have found that protocatechuic acid can inhibit H2O2-induced PC12 cell apoptosis and MPP-induced PC12 cell injury, and its mechanism is related to inhibiting nerve cell apoptosis and improving cell antioxidant capacity. In addition, in vivo animal experiments show that protocatechuic acid can improve endogenous antioxidant enzyme activity in brain tissue, significantly reduce iNOS expression in brain tissue of PD model mice, and at the same time increase TH and D2DR expression, the effect is equivalent to that of the applied drug Madopar for PD. Procatechuic acid preventive administration has obvious effect on the expression of TH, D2DR and iNOS in brain tissue of PD model mice, while the therapeutic administration is stronger. These results show that Procatechuic acid has significant protective effect on nerve injury. |

the Belousov-Zhbaotinskii oscillation system of NaBrO3-MA (malic acid)-H2SO4-[CuL](ClO4) 2 was used as the detection solution to quantitatively analyze protocatechuic acid. Add a series of protocatechuic acid sample solutions with different contents into the oscillation system, and establish a working curve (such as linear diagram) related to the concentration of the measured substance and the change value of the oscillation parameter, so as to achieve the purpose of detecting the content of protocatechuic acid in the sample.
1) prepare solution
2mol/L sulfuric acid solution, 0.5mol/L sodium bromate solution, 2.0mol/L malic acid solution and 1.84 × 10-2mol/L [CuL](ClO4)2 solution were prepared respectively. Then, 20ml, 2.0mol/L sulfuric acid solution, 1.2ml, 0.5mol/L sodium bromate solution, 4.5ml, 2.0mol/L malic acid solution and 8.5ml, 1.84 × 10-2mol/L catalyst solution were successively added to the open system of 50ml beaker. Finally, the concentration of sulfuric acid in the system is 1.0mol/L, the concentration of sodium bromate is 0.0150mol/L, the concentration of malic acid is 0.225mol/L, and the concentration of catalyst is 3.91 × 10-3mol/L.
At the same time, a series of protocatechuic acid sample solutions with different concentrations were prepared.
2) obtain the oscillation pattern
The oscillation map of the prepared oscillation system is recorded with a recorder or computer. A trace amount of protocatechuic acid solution is added to the prepared oscillation solution, and the time of each addition is at the lowest potential of the 7th amplitude generated by stable oscillation. The added protocatechuic acid will participate in the oscillation reaction, that is, the oscillation response will increase the oscillation amplitude.
3) analysis
The working curve is established according to the relationship between the amount of protocatechuic acid added in the system and the logarithmic value of the maximum potential increase. The abscissa is the concentration of protocatechuic acid in the oscillating solution, and the ordinate is the logarithmic value of the highest potential increase value 1g(& Delta;E). When the concentration of protocatechuic acid is between 1.5 × 10-5 and 1.0 × 10-4mol/L, the concentration of protocatechuic acid solution is 1g & Delta;E is a linear relationship. According to this, the quantitative analysis of protocatechuic acid in the sample can be realized.
The reaction steps are:
1) Using vanillin as raw material, 4-hydroxy-3-methoxybenzoic acid was prepared by alkali melting and post-acidification method. The alkali used is strong alkali, preferably sodium hydroxide and potassium hydroxide, the temperature range is 160 ℃-200 ℃, the reaction time is 1-3 hours, and the acid used is hydrochloric acid.
2) Using 4-hydroxy-3-methoxybenzoic acid as an intermediate, it is dispersed in dichloromethane, and then boron tribromide is added to it, and the reaction is in the range of -78 ℃-0 ℃ for 10-17 hours. After the reaction, return to room temperature, then pour it into cold water, extract it with ether, and spin the extract to obtain protocatechuic acid.
Character white to light brown crystalline powder. It turns colorless when exposed to air. Melting point 198~200 ℃ (decomposition). Slightly soluble in water, soluble in ethanol, ether, acetone, ethyl acetate, slightly soluble in benzene, chloroform. Can form salt with alkali. The aqueous solution of ferric chloride is blue-green.
Pharmacological effects have antiasthmatic and antibacterial effects. Protocatechuic acid can effectively improve OVA-induced allergic asthma in mice to a certain extent. Its mechanism may be related to blocking MAPK and NF-& kappa;B, and it is dose-dependent. It has inhibitory effect on Pseudomonas aeruginosa, Escherichia coli, etc. It is extracted from plants and can also be synthesized artificially. It can be used as a fungicide and as a raw material for organic synthesis.
indications have antiasthmatic, expectorant and bacteriostatic effects. For wheezing bronchitis.
