In vitro study | Torsemide inhibits the secretion of aldosterone by murine, bovine, and guinea-pig adrenal cells, stimulated by angiotensin, butyrylcyclic AMP, ACTH, or corticosterone in vitro. The main site of action of Torsemide is the upper Henle circulation of the kidney, which promotes the excretion of sodium, water and chloride by interacting with the Na ,K ,2Cl-transport. Torsemide interferes with mineralocortical aldosterone secretion and binding in a dose-dependent manner. |
In vivo study | In dogs with mitral valve closure, Torsemide causes elevated levels of circulating angiotensin II and aldosterone (which support interference with aldosterone at the receptor level), whereas furosemide only increases levels of angiotensin II. In rats, Torsemide increased blood urea nitrogen and plasma creatinine concentrations compared to baseline values. Torsemide immediately increased urine output significantly. |
