Preparation | 2-chloropyridine-3-sulfonyl chloride can be made of 3-amino-2-chloropyridine as the reaction raw material, it is prepared by diazotization reaction or by the reaction of 2-chloro-3-[(2-chloropyridine-3-yl) dithioalkyl] pyridine with hydrochloric acid and chlorine. Under cold conditions, thionyl chloride (24mL) is added dropwise to water (140mL) within 1 hour, and the mixture is stirred at room temperature for 12 hours to obtain a solution containing sulfur dioxide. Independently, under ice-cold, 3-amino -2-chloropyridine (10g) was added to concentrated hydrochloric acid (80mL) and the mixture was stirred. An aqueous solution (25mL) of sodium nitrite (5.75g) was added dropwise while keeping the internal temperature not higher than 5°C, and the mixture was further stirred for 15 minutes. The reaction mixture is gradually added to the above-mentioned solution containing sulfur dioxide and added with cuprous chloride (140mg) at 5°C. Under ice-cold, the mixture was stirred further for 30 minutes, the precipitate was filtered and washed with water and ethanol to obtain the title compound 2-chloropyridine-3-sulfonyl chloride (yield 6.99g,42%). 1H-NMR(CDCl3)δ:7.54-7.56(1H,m),8.46-8.48(1H,m),8.71-8.73(1H,m). |
