Abstract:
using E. Coli carrying a gene capable of synthesizing poly-3-hydroxybutyric acid. coli XL1-Blue, after optimizing the medium and culture conditions, the Fed-batch culture was carried out. The results showed that the recombinant E. Coli. The optimal medium for coli XL1-Blue(pKSS105) was R medium. Under the optimal conditions, after the engineering bacteria were cultured for 60H with glucose as the sole carbon source, the dry weight of the bacteria in the fermentation broth reached 183g/ L, and the yield of P(3HB) was 133.8g/ L, the P(3HB) content was 73.1%. The experimental results provide a possible basis for the practical application of P(3HB).

Abstract:
objective: to establish an analytical method of D-3-Hydroxybutyric acid automatic biochemical analyzer for monitoring diabetic ketosis. Methods: according to the principle of oxidative dehydrogenation of D-3-Hydroxybutyric acid by D -3-Hydroxybutyrate dehydrogenase in vivo, an in vitro Quantitative determination method was established by enzyme kinetics, to investigate the relationship between D-3-Hydroxybutyric acid and diabetic ketosis. Results: the method is simple, suitable for automatic biochemical analyzer, the experimental indicators to meet the requirements, diabetic ketosis patients D-3-Hydroxybutyric acid content can be as high as 5.12mmoL/L. Conclusion: diabetic ketosis can be detected early by determining the content of D-3-Hydroxybutyric acid.
