Physical and Chemical Properties | Bromothymol blue colorless or light rose crystalline powder, soluble in alcohol, dilute alkali solution and ammonia, slightly soluble in water, insoluble in petroleum ether. When it is acidic, it is yellow; When it is alkaline, it is blue. |
Use | Used as acid-base indicator |
In vitro study | A first characterization and comparison is done by developing an easy and direct measurement method based on a pH indicator system using Bromothymol Blue (BTB) as the indicator and potassium phosphate as the buffer. A pH-shift assay is developed on the basis of BTB as the pH indicator and potassium phosphate as the buffer component. Three pH indicators are tested for the direct determination of 2- 2-keto-L-gulonic acid (2-KLG) production on a plate. The results show that Bromothymol Blue is superior to the other two indicators in terms of the obvious color change and a suitable pH range (blue to yellow at pH 6.5-7.5). Upon the addition of a Bromothymol Blue solution (0.1%, w/v) to an agar plate, zones surrounding colonies of K. vulgare 07 mutants change their color from blue to yellow because K. vulgare 07 mutants release 2-KLG on agar plates, thereby acidifying surrounding areas around colonies. |

Introduction | bromothymol blue is an acid-base indicator with a color change range of pH6.0 (yellow) to 7.6 (blue). Ordinary water is neutral, pH is about 7, almost light green. |
Use | bromothymol blue is a pH indicator of weak acids and weak bases, it is used as an acid-base indicator and a chromatographic analysis reagent in analytical chemistry; It can also be used as a dye and a metabolite. acid-base indicator, pH color change range 6.0 (yellow)-7.6 (blue). Adsorption indicator. |
indicator color change range | pH color change range from 6.0 (yellow) to 7.6 (blue). |
Application | • pH indicator-has precise color conversion at near-neutral, yellow at acidic, and blue at alkaline. • Stain-trace the movement of lymph fluid under a microscope to determine the cell wall or cell nucleus • Medium components-in agar gel medium for Bacillus cereus enumeration; As a bacterial growth component and detection substrate, for example, Cystine/lactose/electrolyte deficiency-BTB disk method-Emergency Determination of whole blood cholinesterase activity for the determination of organophosphate poisoning. |
preparation | Method 1: bromine is introduced into a suspension of Thymol Blue in glacial acetic acid with stirring, the reaction was carried out at 50 °c to obtain bromothymol blue. Method 2: The Excess bromine and glacial acetic acid solution was added slowly to the aqueous acetic acid solution of Thymol blue under stirring, and the reaction was completed at 40~50 ℃ for 4H, it was cooled to below 20 °c, filtered and dried to give colorless bromothymol blue. |
biological activity | Bromothymol Blue is a pH indicator used for the determination of weak acids and bases, when it is acidic, it is yellow, and when it is alkaline, it is blue. |
