In vivo study | In the rat colon, Bisacodyl (20 mg/kg) caused a decrease in AQP3 protein expression and an increase in the mRNA expression level of TNF-α. Bisacodyl inhibited rat jejunum, ileum, and colon water uptake in the range of 0.05 mg to 2.0 mg/100 ml, with a linear correlation to the logarithm of bisacodyl concentration. Bisacodyl (10 mg/kg, gavage) caused a significant decrease in the activity of NOS in rat jejunum. Bisacodyl (10 mg/kg, gavage) increased travel distance for all time period markers. Bisacodyl (5.9 mg/kg) significantly reduced rat jejunum and colon (Na K) ATPase activity compared to saline-treated rats. Bisacodyl (5.9 mg/kg) significantly increased the content of PGE2 in jejunum and colon and stimulated the activity of adenylate cyclase in jejunum and colon compared with control rats which did not affect the content of cAMP.
