CENTRICOLL - Introduction
It is suitable for separating cells, subcellular components and large viruses (>70S), uniform osmotic pressure, low viscosity, adjustable to physiological ionic strength and pH, mild separation conditions, non-toxic to cells, and can be sterilized. Percoll separation solution is a kind of silica gel particles treated with polyvinylpyrrolidone, which can form a continuous density gradient after high-speed centrifugation. Due to the low Percoll diffusion constant, the gradient formed is very stable. When the Percoll separation solution adopts the pre-formed density gradient, satisfactory cell separation results can be achieved within minutes to tens of minutes at low centrifugal force (200~1000g). In addition, the Percoll separation fluid does not penetrate the biofilm and is non-toxic to cells, so it is widely used to separate cells, subcellular components, bacteria and viruses, and can also separate damaged cells and their fragments from intact living cells. Percoll is often used to isolate and tell plant protoplasts, nuclei, chloroplasts, and mitochondria. Configuration and use of Percoll separation solution 1. Preparation of Percoll separation solution with different concentrations (densities): First, 9 parts of Percoll separation solution and 1 part of 8.5% NaCl are mixed to achieve physiological osmotic pressure, and then diluted to the required concentration with physiological solution (0.85% NaCl). 2. Preparation of discontinuous density gradient Percoll layer: First, the test tube wall is moistened with bovine serum to remove excess serum. This pretreatment can make the Percoll liquid superimposed layer by layer flow down the tube wall smoothly to form a satisfactory interface. In the preparation process, a long needle syringe is generally used to place it layer by layer from high density to low density. Sometimes when the specific gravity of the adjacent two layers of Percoll is not much different, the Percoll liquid can be put into the syringe, and the inclined plane of the small needle is close to the tube wall, Let it flow slowly. 3. Sample loading: The sample volume and cell concentration vary according to different cells. Generally, the sample volume should not be too large, and the cell concentration should not be too high, otherwise it will affect the separation and recovery of cells. 4. Centrifugation: Generally, the centrifugal force is 400g for 20~25min. Since the density difference between multi-layer Percoll is not big, the centrifuge should be slow and stable when accelerating and slowing down. 5. Sampling: When most of the cells to be separated are at the interface of two layers, the cells at the interface part can be collected after removing the Percoll solution layer by layer. Sometimes most cells are located in the Percoll layer and need to be collected layer by layer. Cells containing Percoll solution were harvested and washed twice for culture or detection. The advantages of Percoll cell separation fluid Percoll low permeability, no penetration of cells, high density and no toxicity, and is currently an ideal medium. Percoll density gradient centrifugation has been successfully used for many times to separate animal cells and their organelles, and purified more than ten kinds of animal cells including human blood cells, bone marrow cells, red blood cells, white blood cells, lymphocytes, liver cells, etc. Precautions for Percoll cell separation fluid: 1.
