In vitro study | Closantel inhibited drug-resistant Staphylococcus aureus and Enterococcus faecium, MICs 1 μg/mL and 1 μg/mL, respectively. The MRSA MICs inhibited by Closantel was 2 μg/mL. Closantel completely blocks the infection of host cells by E. chaffeensis or A. phagocytophilum, treating in A dose-dependent manner cells persistently infected for one day, thereby clearing the infection. Closantel inhibits the activity of three sensor kinase autokinases from E. chaffeensis. Closantel (50 μg/mL) acts on vascular tissue to induce contractions of greater amplitude and higher frequency than normal. Stimulation at such amplitude and frequency for about 15 minutes is accompanied by an increase in muscle tension, this results in a maximum of 16 min, which is a level greater than 1.5 times the maximum normal amplitude. |
In vivo study | Closantel (10 mg/kg) acted on 25 F before administration. Mice with posterior metacercaria of the hepatica family, after 24 hours, cause total surface damage in mice in the form of erosion of the anterior and posterior limbs of the fluke and extensive molting on the back and abdominal surfaces. Closantel (7.5 mg/kg) in combination with a broad-spectrum anthelmintic was very effective against the nematode twister, but not very effective against the genus Strongylus in sheep. Closantel significantly reduced isotope levels in closantel-resistant, adult sheep infected with the twisted blood spear nematode. |

Toxicity | A veterinary drug for the treatment of fluke and nematode infestations. In 1993, some was included in a batch of donated drugs sent to Lithuania. Attempts to match the product with the names of other medicines on accompanying literature resulted in its misidentification as a treatment for endometriosis. Eleven women lost their eyesight; though this has since returned, they were left with eye pain and are unable to work. |
