| preparation method | one equivalent of N α-fluorenylmethyloxycarbonyl-N ε-tert-butoxycarbonyl-L-lysine was dissolved in dichloromethane, add 1 part of dichloromethane solution of 15% trifluoroacetic acid by mass, stir vigorously, cool externally with tap water, check the reaction completely by TLC, distill off the solvent and trifluoroacetic acid under reduced pressure, add dichloromethane to dissolve again, again under reduced pressure distillation, petroleum ether washing, petroleum ether poured off, the remaining solid dissolved in dimethylformamide, add 2 equivalents of N-methyl morpholine, pH value is adjusted to 8, slowly add 0.7 equivalent of N-hydroxysuccinimide biotin, react for 3 hours, thin layer chromatography showed complete reaction, add citric acid solution to the reaction, pH adjusted to 3, solid precipitation, washing, drying, the product was obtained with a purity of 99.1 and a yield of 90%. |
