preparation | take a 500mL reaction flask and add chiral amino acid S-3a(4.5g,38.8mmol), dioxane (40ml) and 10% sodium carbonate (10.0 ml), the reaction flask was placed in an ice bath, mechanically stirred, and to the dropping funnel was added 9-fluorenyl methyl chloroformate (38.8g, mmol). And dioxane (100ml), slowly dropped into the reaction flask, gradually returned to room temperature and stirred overnight. After completion of the reaction, 100ml of water was added and extracted three times with 50ml of ether. The aqueous phase was cooled in an ice bath and 1m of dilute HCl was added to PH 1. The aqueous solution was extracted three times with 50ml of ethyl acetate. The oil phases were combined and dried over magnesium sulfate. Intermediate S-4a, I .e., Fmoc-L-valine (12.6g,96%), was obtained after filtration and spin-drying. White solid. |
