7-guanidinylheptanamide (I,22.3g,100 mmo1) and glyoxylic acid (10.2g,110 mmo1) are dissolved in 150ml of dimethylformamide and reacted at 65~70 ℃ for 4h. Cooling, adding 110ml benzyl alcohol and 6ml concentrated hydrochloric acid, stirring for 42h. Use 6mol/L hydrochloric acid to adjust the Ph value to 12.0, and then stir at room temperature for 40min. Use 6mol/L hydrochloric acid to adjust the Ph value to 7.0, and then dilute with 1L water. The precipitate was filtered out, the filtrate was purified by Diaion SP-207 column, and eluted with 50% ~ 60% methanol aqueous solution. The eluent containing the product was collected and concentrated. The residue was crystallized with methanol aqueous solution to obtain 9.45g compound (II) with 27% yield.
Compound (II)(7.92g,22.6 mmo1) is dissolved in 30ml methanol and 1mol/L hydrogen chloride methanol solution, concentrated under reduced pressure and dried to obtain hydrochloride of compound (II). Dissolve the hydrochloride (7.9g,20.4 mmo1) in 80ml dimethylformamide, add N-hydroxysuccinimide (3.12g,27.lmmo1) and DCC(5.60g,27.1 mmo1), and stir at room temperature overnight. Add L-arginine (4.72g,27.1 mmo1) and triethylamine (3.8ml,27.1 mmo1) in a 40ml water solution and stir for 7h at room temperature. Add 100ml of water, adjust to Ph value 12 with 6mol/L sodium hydroxide, and stir for 1h. The precipitate was filtered out, and the filtrate was purified by Diaion HP-20 column and eluted with 30% methanol to obtain 5.41g compound (Ⅲ) with 44% yield.
