In vitro study | At low concentrations (nM),INCB057643 inhibited binding of BRD2/BRD3/BRD4 to acetylated histone H4 with selectivity. In in vitro experiments, INCB057643 inhibits proliferation of human AML, DLBCL and multiple myeloma cell lines with reduced levels of MYC protein. Treatment of cells with INCB057643 for 48 hours induced G1 phase arrest and a concentration-dependent increase in apoptosis. In human and mouse whole blood cells, INCB057643 inhibited LPS-stimulated production of cytokines, including IL-6, IL-10 and MIP-1α. INCB057643 inhibits the proliferation of prostate cancer cells. In the short-term cell proliferation assay, INCB057643 was more effective on androgen-dependent cells (VCaP and LNCaP) than on non-androgen-dependent cells (DU145 and PC3). |
In vivo study | In AML, DLBCL and myeloma transplantation models, oral INCB057643 caused significant anti-tumor effects. In the mouse model inoculated with 22Rv1 tumor cells, 3 mg/kg significantly inhibited tumor growth (T/C%:45%) and sustained reduction in tumor weight. |
