biological activity | analysis of the expression level of alloxins on MMP-1: HaCaT cells were treated with alloxins for 24 hours, and irradiated with 20MJ/cm2 of ultraviolet light (UV crosslinker, UltraLum), then using the MMP-1ELISA kit (R & D Systems,Inc., Minneapolis, MN, USA) the treatment was performed and the expression level of MMP-1 was measured. Specifically, HaCaT cells were plated on a 96-well plate, and then treated with different concentrations of different concentrations of yellow glycosides. In addition, as a comparative example, the extract, water component, ethyl acetate component, butanol component, the hydroxysteviol glycosides were concentrated and then subjected to ultraviolet (UV) irradiation. The culture supernatant was collected and centrifuged at 13,000rpm for 5 minutes. The supernatant was then taken and subjected to a fluorescence assay using an ELISA kit to quantify the expression level of MMP-1. The results showed a significant increase in the expression level of MMP-1 in the UV-irradiated group compared to the negative control group and the experimental group without UV irradiation confirmed that compared to the UV-irradiated group, the expression level of MMP-1 was significantly reduced in the cells treated with hetero-xanthine. |
