preparation | The Flowers of Pueraria lobata were crushed and weighed. 300g was extracted three times with 900 mL of methanol, and the combined extracts were concentrated under reduced pressure. The methanol extract was suspended in an appropriate amount of distilled water, defatted with benzene extraction, then extracted with n-butanol, the n-butanol extract was concentrated to dryness, and then eluted by silica gel column chromatography with CHCl3: CH3OH (10: 1), light yellow, snow-like crystals were obtained (re-crystallized from methanol), I .e., Pueraria glycosides were obtained. |
Method for Determination of content | UV-Spectrophotometry: the content of isoflavones in Pueraria lobata was determined by UV-spectrophotometry. The UV detection wavelength was 265mm, there was a good linear relationship in the range of 0.01-0.04mg/L. The precision, sensitivity and stability of UV method for the determination of Pueraria glycosides in Pueraria lobata flower were good, fast, suitable for the determination of the concentration of Pueraria in the production process. Reversed-phase high performance liquid chromatography: a LUNA-C18( 250mm × 4.6, 5um) column was used with the mobile phase of acetonitrile-0.1% phosphoric acid solution (18: 82) at a flow rate of 1. 0 ml/min, column temperature 30 C, detection wavelength 265 nm. There was a good linear relationship between the peak area and the range of 0.062~1.240 mg /mL. The method was accurate, reliable, sensitive and specific. dual-wavelength thin-layer scanning method: a dual-wavelength thin-layer scanning method was developed for the determination of Flos puerariae glycosides in Pueraria lobata. The developing solvent of TLC analysis was chloroform-methanol-glacial acetic acid (3 ∶ 1 ∶ 0.3), and the glycoside of Pueraria showed good linearity in the range of 0.484~3.872 μg. The method is rapid and simple, and provides a scientific basis for the quality control of Pueraria lobata as traditional Chinese medicine. |
