In vitro study | Methylclothiazide (0.1 mM) reduces Ca methylclothiazide (0.1 mM) Intima-dependent inhibition of the vasoconstrictive response of the aorta to NE and AVP in spontaneously hypertensive rats (SHR), relative to normotensive Wistar Kyoto rats (WKY), the maximal vasoconstrictor effect of NE and AVP decreased by 59% and 32.3, respectively. Methyclothiazide (3.5 mM) inhibits renin release in rat kidney slices. |
In vivo study | Methylclothiazide (0.1g/L for the first 5 weeks and 0.2g/L for the next 2 weeks in drinking water) attenuates the development of hypertension in salt-sensitive (DS) rats, but did not affect the blood pressure of salt-tolerant (DR) rats. At Weeks 3 and 4, methylclothiazide-treated DOCA-salt rats had lower body weights than control or unsupplemented DOCA-salt rats. Methylclothiazide was added to the drinking water of DOCA-salt rats to attenuate the increase in blood pressure. |
