In vitro study | Mirtazapine exhibits remarkable affinity for the cloned human α2a-adrenaline (AR) receptor, at which it blocks noradrenaline (NA) induced guanosine-5 '-O-(3-[ Mirtazapine enhances electrical stimulation of the ascending 5-HT pathway by blocking α2 Adrenergic self-and hetero-receptors. Mirtazapine blocks the inhibition of the firing activity of CA3 dorsal hippocampal vertebral neurons by norepinephrine (NE) applied by microparticle electroosmosis, suggesting an antagonistic effect on postsynaptic α2 receptors. |
In vivo study | Mirtazapine (10-250 mg/kg I. v.) dose-dependently enhanced the firing activity of 5-HT neurons in naïve rats but had no effect on rats pretreated with 6-hydroxydopamine. In male Sprague-Dawley rats, Mirtazapine (5 mg/kg/day, S. C., using a subcutaneously embedded osmotic micropump) increased the spontaneous firing activity of norepinephrine (NA) neurons in the locus coereurus. For the inhibition of the discharge activity of CA3 pyramidal neurons in the dorsal hippocampus, the α2 adrenoceptor agonist Clonidine has an effect on the ascending 5-HT pathway electrical stimulation, Mirtazapine at a low dose (10 mg/kg, I. v.) under antagonistic potentiation, at high doses (100 mg/kg, I. v.) under the antagonistic weakening effect. Mirtazapine (5 mg/kg s.c.) only slightly affected the levels of DOPAC and homovanillic acid in the striatum and hardly affected the release of 5-HT in freely moving rats, but significantly increased 5-hydroxyindoleacetic acid. |
