preparation | Na2CO3 (8.9g, 84 mmol) was added to 1N NaOH (168 mL) solution of L-alanine (8g, 168 mmol). The solution was cooled to 0°C and methyl chloroformate (20.5 mL, 172 mmol) was added dropwise. The reaction mixture was stirred at room temperature for 3.5 hours. The solution was washed with Et2O and acidified with 1N HCl to a pH of about 1. The water layer was extracted with DCM, the organic layer was washed with brine, dried with Na2SO4, and concentrated to obtain the target intermediate N-methoxycarbonyl-L-tert-leucine. |
