Introduction | 2-amino -4, 6-dichloro-5-formamidopyrimidine is an important intermediate of abacavir. Abacavir is a new anti-AIDS drug, a new carbocyclic 2 '-deoxyguanosine nucleoside drug, its oral bioavailability is high, easy to penetrate into the central nervous system. The drug is a nucleoside reverse transcriptase preparation obtained by Glaxo Wellcome through the FDA rapid approval process in December 1998. In April 2004, it was approved by the US FDA for use against HIV infection. |

preparation | adding 146.0g(2.0mol) of nitroso-formidine, 584.0g of sodium methoxide and 240.0g of formamidomalononitrile in example 2 into a 1000mL four-port reaction bottle, raising the temperature to reflux reaction, evaporating methanol under reduced pressure to separate the solid, slowly cooling to 5 ℃, filtering and drying to obtain 353.0g of solid, the purity of HPLC was 99.5% and the yield was 97.0%. Preparation of 2-Amino -4,6-Dichloro -5-formamido pyrimidine (VI) 100.0g(0.45mol) of 2-nitroso -4,6-Dichloro -5-formamido pyrimidine (mol), 400.0g of methanol and 8.0g of 5% palladium and carbon were added into a 1L autoclave, and hydrogen was replaced by hydrogen to maintain 1.0-1.5MPa, the temperature is kept at 40 ℃ for 4 hours. After the detection reaction is completed, filtration (filter cake is recycled and applied), methanol is evaporated from the filtrate, water is added to cool down to 5 ℃, filtration and drying are performed to obtain 91.3g of solid. The purity measured by HPLC is 99.4% and the yield is 98.0%. |
