Summary:
objective: to study the effect of pine needle oil on human hepatoma HepG2 cells. Methods: pine needle oil was extracted by steam distillation, and its composition was GC-MS analyzed. After different concentrations of pine needle oil were treated with HepG2 cells, the growth inhibition rate of HepG2 cells was determined by MTT method. Morphological changes of apoptosis were observed under HE staining and electron microscope. Flow cytometry was used to detect apoptosis rate. Flow cytometry was used to examine the expression level of bcl-2 genes. TRAP method was used to determine the changes of telomerase activity. Results: MTT method proved that pine needle oil can significantly inhibit the growth of HepG2 cells, and the inhibition rate is positively correlated with the drug concentration, showing obvious dose-effect and time-effect relationship. Apoptosis can be detected by flow cytometry, and 0.125~8 mg/L pine needle oil can cause HepG2 cell apoptosis in a time-dependent and dose-dependent manner. Typical apoptosis characteristics such as nuclear concentration and nuclear fragmentation can be observed under electron microscope. Pine needle oil induces apoptosis and decreases the expression level of telomerase activity and bcl-2. Conclusion: pine needle oil induces apoptosis of hepatoma HepG2 cells and inhibits telomerase activity, which bcl-2 participates in the regulation of telomerase activity during apoptosis of hepatoma HepG2 cells induced by pine needle oil.

Summary:
using gelatin/modified gum arabic as wall material and pine needle oil as core material, pine needle oil microcapsules were prepared by composite coagulation method. The optimum reaction conditions for preparing pine needle oil microcapsules are as follows: gelatin/modified Arabic gum ratio is 1.25:1, mass fraction is 0.6%,pH = 4.1, reaction time is 75min, stirring speed is 300rpm, curing temperature is 45 ℃, curing time is 10h. Optical microscope (OM), scanning electron microscope (SEM), Fourier transform infrared spectrometer (FTIR) and thermogravimetric analyzer (DTG-TA) were used to characterize the morphology and thermal properties of pine needle oil microcapsules. The results showed that the prepared pine needle oil microcapsules had uniform particle distribution and good thermal stability and slow release properties. The fabric finished by pine needle oil microcapsules shows good slow release performance and water washing resistance.
