In vitro study | RTA408 effectively increased the expression of Nrf2 target genes in RAW 264.7 cells and reversed IFNγ-mediated inhibition of Gclc expression. In a panel of 8 human tumor cell lines, RTA 408 inhibited growth with a mean GI50 value of 260 nM and induced apoptosis. In tumor cells, RTA 408 also inhibits NF-κB and activates JNK. |
In vivo study | In radiation-induced dermatitis mice, 1.0% RTA 408 significantly reduced epidermal and collagen thickness, prevented dermal necrosis, and completely alleviated skin ulcers. In rat skin, RTA 408 activates Nrf2 and induces cytoprotective genes. RTA 408 also reduced hematopoietic acute radiation syndrome in mice. |
