Summary:
Mung Bean nuclease (Mung Bean Nuclease) can cut off the genomic DNA of malaria protozoa Plasmodium before and after the gene but not in the coding region of the gene. This cutting has almost the accuracy of restriction endonuclease and requires formamide control. Southern blotting showed that the gene coding regions of actin, CS protein (circum-sporozoite protein), histidine-rich egg, ribosomal RNA-binding protein and tubulin of Plasmodium were all cut from genomic DNA one by one to form a single narrow band on agarose gel. Summary:
objective] to optimize the extraction conditions of mung bean nuclease and determine its activity. [method] single factor and orthogonal test were used to investigate the effects of extraction time, solid-liquid ratio, pH and bud length on the extraction of nuclease from mung bean sprouts. the optimal level of each influencing factor was optimized and the activity was determined. [results] the extraction time was 20 h and the solid-liquid ratio was 1:12: mL),pH 3.0, bud length is 4 times that of mung bean, mung bean nuclease activity is 478.86U/ mL. The main order of factors affecting the extraction of mung bean nuclease is extraction time, pH, solid-liquid ratio and bud length. After ammonium sulfate with saturation of 80% was salted out and precipitated, the enzyme activity reached 1 569.6U/ mL, which was 2.28 times higher than that before purification. [conclusion] the study can provide reference for the development and utilization of nuclease.
