In vitro study | Inhibition of DPP4 by Saxagliptin has an inhibition constant Ki of 1.3 nM, which is 10-fold more potent than that of villagcliptin or sitagliptin (two other DPP4 inhibitors) with K I of 13 and 18 nM, respectively. In addition, Saxagliptin was more specific for DPP4 than DPP8 or DPP9 enzymes (400-and 75-fold, respectively). The active metabolite of Saxagliptin is 2 times less potent than Saxagliptin. Compared to a range of other proteases (> 2600 and <250-fold selectivity for DPP4 for sitagliptin and villagliptin over DPP8 and DPP9, respectively), both Saxagliptin and its active metabolite were highly selective (>4000-fold) for inhibition of dpp4. Saxagliptin reduces the degradation of the intestinal hypoglycemic hormone glucagon-like polypeptide -1, thereby enhancing its effect, and is associated with improved beta cell function and inhibition of glucagon secretion. |
