In vivo study | In nephrectomized rats fed a high-phosphorus (HP) diet, Sevelamer was as effective as calcium carbonate, reduced serum phosphorus, calcium-phosphorus product, and slowed secondary hyperparathyroidism. Compared to control mice, Sevelamer was able to resolve leading to a significant reduction in calcium deposition in the myocardium and aorta. In rats with chronic renal failure, Sevelamer inhibited aortic calcification and also inhibited osteoid volume, fibrotic volume, and femoral porosity ratio. Sevelamer significantly reduced atherosclerosis and vascular calcification in uremic mice compared to uremic controls. In uremic mice, Sevelamer had an effect on both intimal and medial calcification. In uremic mice, Sevelamer treatment controls the increase in serum calcium independent of serum phosphorus, thereby reducing serum calcium and phosphorus and further deterioration of renal function, expressed as maximum creatinine clearance. Sevelamer is as effective as calcium carbonate in inhibiting high P-induced SH, as marked by similar serum PTH levels, parathyroid gland weight (PT) and PT hyperplasia. Sevelamer resulted in a significant reduction in renal calcium deposition compared to the uremic high-P diet (U-HP) and the U-HP calcium carbonate diet. In uremic mice, Sevelamer hydrochloride caused a decrease in urinary pH and an increase in urinary ammonium and calcium excretion, consistent with net acid excretion in animal models. |
