Abstract:
An ultra performance liquid chromatography-triple quadrupole mass spectrometry (UHPLC-MS/MS) method was developed for the determination of methacholine chloride aerosol in inhalation devices. The chromatographic column was an acquisition BEH C_(18)(2.1 × 100, 1.7 μm) column, and 0.1% formic acid water and acetonitrile were used as the mobile phase for gradient elution at a flow rate of 0.3 mL/min, the sample volume was 5 μL, the analysis time was 5 min, the column temperature was 40 ℃, and the multiple reaction monitoring (MRM) positive ion mode was determined. At the same time, the specificity, linearity, instrument precision, standard recovery rate, extraction recovery rate and stability were investigated. The results showed that the calibration curve of methacholine chloride was linear within the range of 10~100 ng/mL (r> 0.999), and the limit of quantitation was 10 ng/mL; the average recovery of methacholine chloride aerosol was 99.2%, and the average relative standard deviation (RSD) was 0.80%. The average recovery of methacholine chloride aerosol was 95.7%. After nebulization in the inhalation exposure device for 1 min, the average drug concentration of the aerosol was 608.1 mg/m ^(3), and the RSD of the upper, middle and lower positions was 1.2%, the results showed that the aerosol concentration of methacholine chloride was uniform within 1 min of nebulization. The method has high sensitivity and good precision, which is suitable for the concentration determination of methacholine chloride aerosol.
