In vitro study | Hexamethonium Bromide was effective against Ach and Carbachol (CCh) responses to the amplitude of the rat scaphohyoid muscle endplate, with EC50 of 300 μm and 100 μm, respectively. The amplitude of neuro-evoked endplate currents (e.p.cs) was increased by Hexamethonium Bromide (50-200 μm) in the presence of 0.6 μm tubocurarine. In rat muscle homogenates, Hexamethonium Bromide was also a weak inhibitor of acetylcholinesterase activity with an EC50 of 1.5 mM. In rat diaphragmatic muscle on one side, Hexamethonium Bromide (200 μm) decreased endplate currents (e.p.cs) (reduced by ~ 25%) and micro-endplate current (m.e.p.cs) (reduced by ~ 20%) decay time constant. Under low-frequency stimulation (0.5-2Hz), Hexamethonium Bromide (200 μm) increases the quantum content of e.p.c. By 30-40%. |
