In vitro study | Adrenalone (12 μm) inhibits the conversion of dopamine to norepinephrine by inhibiting dopamine beta oxidase. The structure of Adrenalone differs from that of Epinine, which has a ketone group at the β site. Adrenalone (0.1 mM) in a nitrided aqueous solution containing 10 or 100 mM of tert-butanol, decreased Adrenalone by hydrating electrons, resulting in the same transient spectrum, so that all the OH radicals were scavenged. Although Adrenalone acts similarly to carbonyl compounds in the reduction process, it is mainly controlled by the catechol functional group in the oxidation reaction. Adrenalone is a topical nasal vasoconstrictor, hemostatic, vasoconstrictor, a keto form of the natural substrate epinephrine. Adrenalone contains an aromatic ring (forming hydrophobic interactions with Phe72,Tyr152, and Phe317), an amine group (forming an ionic bond with Asp75), and a hydroxyl group (forming a hydrogen bond with Ala145). 10 μm (100 μm) Adrenalone reduced substrate uptake by 99%(27%). |
In vivo study | Adrenalone (0.05%) was administered externally to the eyes of rabbits at 30-minute intervals, and the concentrations of cornea, aqueous humor and iris-ciliary body were 7.75 mg/kg, respectively, 0.87 mg/kg and 2.51 mg/kg. |
