p-methoxyphenylacetonitrile was dissolved in Tetrahydrofuran, and a solution of lithium butane in hexane was added under nitrogen. Further, cyclohexanone was added to perform the reaction. A saturated aqueous solution of ammonium chloride containing ice was added to the reaction solution, and further water was added. The precipitate was filtered, washed with water and ether to give 1-[cyano (4-methoxyphenyl) methyl] cyclohexanol. It was dissolved in ammonia-ethanol mixture, and germanium-alumina was added for catalytic hydrogenation. The catalyst was filtered off and washed with ethanol. The washings and filtrates were combined and concentrated to an oil. The oil, aqueous formaldehyde solution, formic acid and water were stirred together overnight. The reaction solution was concentrated, water was added, adjusted to pH 2 with concentrated hydrochloric acid, and extracted with ethyl acetate to remove pink impurities. The extracted aqueous solution was basified with sodium hydroxide solution and then extracted with ethyl acetate. The extract was washed with saturated brine, dried, filtered and concentrated. The resulting material was dissolved in ethyl acetate, a solution of hydrogen chloride in isopropanol was added, and venlafaxine hydrochloride was collected. The racemic venlafaxine was dissolved in ethyl acetate, and the ethyl acetate solution of Di (p-tolyl)- L-tartaric acid monohydrate was added to separate to obtain () one or (one) of the optical isomer.
