| prepare | 60g crude extract of hawthorn leaves, add 500mL distilled water for ultrasonic dissolution, filter out insoluble matter, and flow through HPD-400 macroporous resin column at a flow rate of 3BV/h; After the adsorption is completed, the HPD-400 macroporous resin column is fully eluted with 5BV distilled water to collect the first stage eluent, after concentration and freeze-drying, 5.49g of chlorogenic acid compound with a purity of 70.3% was obtained. HPD -400 macroporous resin column was fully eluted with 5BV 50% ethanol aqueous solution, the second-stage eluent was collected, and 19.32g of hawthorn leaf total flavonoids with a purity of 59.7% were obtained after decompression distillation and recovery of solvent. The total flavonoids of hawthorn leaves are dissolved in 50mL methanol and refined by preparative chromatography {C18 column (50 '250mm,10 μm),35/65(V/V) 0.1% acetic acid aqueous solution/methanol}, the monomer eluent was collected respectively, and after distillation under reduced pressure, 0.29g of vitexin glucoside with purity of 96.2% and 1.29g of vitexin rhamnoside with purity of 96.3% were obtained. |
