Caution | The cations in toluidine blue bind to the acidic substance of the tissue cells and are stained. The staining results showed that the nucleus was blue, while the cytoplasm of mast cells contained hepar |
Use | Toluidine Blue (Toluidine Blue O) is a commonly used synthetic dye, belonging to the class of quinonimine dyes, this class of dyes mainly contains two chromophores, amine groups and quinone-type benzene rings, thereby developing the chromogen. Toluidine blue staining solution (cell-specific) is suitable for intracellular acid mucopolysaccharide staining, especially for sulfur-containing acid mucilage staining, acid mucopolysaccharide staining red. |
staining principle | The cation in Toluidine blue has a staining effect, and the acidic substance of the tissue cells is stained in combination with the cation in it. Toluidine blue also contains two chromophores, which can promote the ionization of the dye into salts, help the chromophore to produce staining force on the tissue, make the tissue cells on the section stain, can stain the nucleus to make it blue. In addition, the cytoplasm of mast cells contains heparin and histamine and other heterochromic substances. When encountered, Toluidine blue can be colored purple, which is often used for mast cell staining in clinic. |
Procedure | 1. Immediately after the smear, the cells were placed in 95% ethanol for fixation for 15s, then removed and placed on a tissue. 2. Toluidine blue staining solution (special for cells) was added dropwise for 5min. 3. Add equal amount of distilled water Dropwise and mix on the smear, stain for 15min. 4. Washing, drying and microscopic examination were performed. staining results: acidic mucopolysaccharide in cells was red. Note: to confirm the staining effect, chronic myeloid leukemia blood smear (eosinophils) or aplastic anemia bone marrow smear (tissue eosinophils) can be used. As a control. for your safety and health, please wear lab coat and disposable gloves. |
