酪蛋白异硫氰酸荧光素来源于牛奶 - Introduction
Casein fluorescein isothiocyanate is a naturally occurring fluorescein in milk. It is a protein formed by the covalent binding of a casein molecule and isothiocyanate phenylase (FITC).
Casein fluorescein isothiocyanate has strong fluorescence properties and can emit green fluorescence under long-wave ultraviolet excitation. It is widely used in biology and biochemistry because of its stable spectral properties, high fluorescence quantum yield and fluorescence lifetime. It can be used to label biological macromolecules such as proteins, cell membranes, organelles and nuclei, making them visible under the microscope, and to locate, track and study the behavior and interaction of biological molecules.
The method of preparing casein fluorescein isothiocyanate is generally by reacting casein with isothiocyanate phenylase (FITC) under alkaline conditions to covalently bind the two. The reaction is usually carried out at room temperature, and purification and centrifugation are carried out after the reaction is completed to obtain purified fluorescein.
casein fluorescein isothiocyanate needs to be safe during use. It is less toxic to humans, but care is still needed to avoid contact with the mouth, eyes and skin. Use appropriate protective equipment such as gloves and goggles. At the same time, light and high temperature environment should be avoided during storage to maintain its stability and fluorescence performance. In laboratory operations, the relevant laboratory safety specifications and operating procedures should be strictly observed.
