Summary:
Objective To establish a simple, rapid and accurate method for the determination of uronic acid in Tremella polysaccharide. Methods The interference of neutral sugar in the determination of uronic acid in carbazole method and m-hydroxybiphenyl method was investigated, and the inhibitory effect of sulfamic acid on neutral sugar was investigated. The uronic acid in Tremella polysaccharide was determined. Results Neutral sugars were interfered to different degrees in carbazole method and m-hydroxybiphenyl method. When the concentration of acidic heteropolysaccharide in uronic acid exceeds a certain range, the results of uronic acid measured by the two methods will have a large error with the actual value. Conclusion the determination of tremella polysaccharide by m-hydroxybiphenyl method, and the sample mass concentration is prepared at 0.25~0.50 mg/mL, the measured results are closer to the actual value.

Summary:
the method of obtaining tremella polysaccharide by hot water extraction, ethanol precipitation and sevag method is discussed. In the reaction of in vitro chemical simulation system, the polysaccharide was observed to eliminate hydroxyl radical, superoxide anion radical and prevent lipid oxidation. The test shows that Tremella polysaccharide has obvious scavenging effect on hydroxyl radical, the scavenging amount of 50% is 0.112 mg/ml, the scavenging ability of superoxide radical increases with the concentration of polysaccharide, and the 50% scavenging amount is 0.264mg/m1. The antioxidant properties of oils are between VE and VC.

TREMELLA - Study on Extraction Technology of Tremella Polysaccharide

Summary:
various extraction methods of polysaccharide from tremella fruiting bodies were discussed and the extraction method of combined alkali extraction under ultrasonic enhancement was studied by orthogonal test. The results showed that the extraction method of alkali extraction after ultrasonic treatment could significantly improve the extraction rate of polysaccharide in the extract. the optimal extraction process conditions of tremella polysaccharide were hot water extraction for 20h, then ultrasonic treatment for 8min, ultrasonic power 400W, alkali concentration of residue alkali extraction for 10%, and extraction yield of 31.77%.
