In vitro study | Butyrate mediates growth inhibition of colon cancer cells, thereby elucidating the molecular link between a high-fiber diet and suppression of colon carcinogenesis. The Butyrate induces the expression of p21 mRNA in an immediate early manner, through the 1.4KB promoter cis-element (s) located at the transcription start site, independent of p53 binding transactivation. Butyrate induced apoptosis in 2 adenoma cell lines RG/C2 and AA/Cl and cancer cell line PC/JW/FI at physiological concentrations. Butyrate has powerful effects on a variety of colonic mucosa, such as inhibiting inflammation and tumorigenesis, strengthening various components of the colonic defense barrier and reducing oxidative stress. Butyrate inhibited most HDAC except Class III HDAC and class II HDAC6 and -10. Butyrate is also the most effective fatty acid to stimulate or inhibit the expression of specific genes. |
In vivo study | In a dose-dependent manner, Butyrate sodium significantly prolonged survival, increased body weight and exercise capacity, and delayed the pathological sequelae in an R6 /2 transgenic mouse model of Huntington's disease (HD). Butyrate sodium salt also increases histone and specific protein -1 acetylation and protects against 3-nitropropionic neurotoxicity. |
