Uses | 2,4-dibromothiophene is an organic intermediate that can be prepared from thiophene through a two-step reaction. It has been reported in the literature that it can be used to prepare ERR inverse agonists. |
preparation | 2,3, 5-tribromothiophene (1):120g (1.43 mol) of thiophene is added with 240 ml of glacial acetic acid, 687g (4.30 mol) of Br2 is added dropwise at 5-10 ℃, stirring at room temperature for 3 hours and refluxing heating for 2 hours. Subsequently, glacial acetic acid was evaporated under vacuum and the residue was distilled through a short Vigreux column under water jet vacuum. Yield: 376.2g(1.17mol)1(82%). 2,4-dibromothiophene (4): 120g(0.374mol)2,3, 5-tribromothiophene (1) is dissolved in 300mL of diethyl ether, cooled to -30 ℃ under N2, and at this temperature, 234mL6NBuLi n-hexane solution (0.374mol) is added dropwise within 10 minutes, stirred at -30 ℃ for 5 minutes, 1L of cold water is poured, and extracted with diethyl ether 3 times. The combined ether phase was washed 3 times with water, dried with Na2SO4 and evaporated. The residue was fractionated on a 1m Vigreux flask under vacuum. Yield: 60g(0.248mol)4(66%). |
