In vitro study | Nordihydroguaiaretic acid (NDGA) (< 1.5 μm) decreased the sister chromatid exchange rate (SCEs) in human lymphocytes treated with 150 μg/mL ethynodiol diacetate. NDGA treatment in the presence of S9 mixture will reduce the genotoxic damage caused by ethynodiol diacetate, this may be achieved by inhibition of cytochrome P450s, which prevents the metabolic activation of ethynodiol diacetate, or by NDGA's free radical scavenging function. Ethynodiol diacetate (150 μg/mL) increases sister chromatid exchange rate (SCEs) and chromosomal aberrations (CAs) in human lymphocytes in the presence of S9 mixture frequency and can inhibit lymphocyte proliferation. |
In vivo study | Hynodiol diacetate at a dose of 6 mg/kg per day can significantly increase the content of A- homo-3-oxa-5alpha-pregnane-4 and 20-dione in the urine of female rats, and can also significantly increase the weight of the pituitary gland, liver and kidney of rats. Combination of Norgestrel (75 μg/day) with ethynodiol diacetate (150 μg/day) and ethinyl estradiol (7.5 μg/day) reduces plasma concentrations of high-density lipoprotein cholesterol in cynomolgus monkeys |
