Abstract:
endophytes are microorganisms living in healthy plant tissues. Studies have shown that the endophyte of traditional Chinese medicine can produce abundant metabolites, including the same or different chemical components with the host. Cordyceps sinensis is a genus of ergot family Cordyceps fungus Cordyceps sinensis Cordyceps sinensis Sacc. A complex of larvae and larvae that are parasitic on the larvae of the hepiales armicanus Oberthuk insect.
It is reported that there are many kinds of endophytes in Cordyceps sinensis, and the secondary metabolites produced by some endophytes isolated from them show different biological activities. In order to search for compounds with novel structure and high activity, we isolated endophytes from fresh Cordyceps sinensis and studied the chemical constituents of its secondary metabolites. In order to make the endophytic fungus of Cordyceps sinensis produce abundant secondary metabolites under laboratory culture conditions, thus increasing the possibility of producing bioactive chemical components.
five epigenetic modifiers, 5-AC, decitabine, SAHA, sodium butyrate, and valproic acid, were added to the fermentation broth to stimulate silenced gene expression. The fermentation broth to which the modifier was added and the fermentation broth to which the modifier was not added were extracted with ethyl acetate, and the extracts were subjected to HPLC analysis to compare the HPLC profiles with and without the modifier. The strains with abundant metabolites, relatively more species, moderate polarity, obvious and stable changes in HPLC profiles with and without modifier were selected as the follow-up study objects.
we selected a strain of endophytic fungus Aspergillus fumigatus Aspergillus fumigatus, added SAHA in the fermentation broth, expand the fermentation, the fermentation broth was extracted with ethyl acetate, and the chemical composition of the extract was studied, using normal phase silica gel, sephadex gel sephadex LH-20 column chromatography and preparative and semi-preparative chromatography for separation and purification, the obtained compounds were identified by spectral analysis. includes 12 polyketones, 10 diketopiperazines, 6 sesquiterpenes, 2 acetanilides, and 2 others. Among them, 16 new compounds (1a ~ 13), including 5 sesquiterpenes (9~13),11 polyketones (1a ~ 8). The cytotoxic activity of some new compounds was tested, and it was found that the new compounds 1a,2 had weak cytotoxic activity against MV4-11 (Human acute leukemia cell line); Compound 5 on H1299 (human lung cancer cells), h460 (large cell lung cancer cells),HGC-27 (human gastric cancer cells),A549 (human non-small cell lung cancer cells) and mnk-43 (human gastric cancer well differentiated cells) the five cell lines had weak cytotoxic activity.
