In vitro study | 5-FAM contains a carboxylic acid that can be used to react with primary amines via carbodiimide activation of the carboxylic acid. Fluorescein is the most common fluorescent derivatization reagent for labeling biomolecules. In addition to its relatively high absorptivity, excellent fluorescence quantum yield, and good water solubility, fluorescein has an excitation maximum that closely matches the 488 nm spectral line of the argon-ion laser. |

Introduction | 5-carboxyfluorescein is a useful reagent for the preparation of hydrolytically stable fluorescent conjugates, it is also a useful starting material for the synthesis of other fluorescein-derived reagents. It is a mixture of 5-and 6-isomers prepared by the reaction of benzenetricarboxylic anhydride with resorcinol and zinc chloride. Although these isomers have almost the same properties, when bound to other molecules, in the case of conjugated polarity, internal fluorescence quenching, and related Rhodamine-labeled reagents, differences in marker specificity have been observed. In addition, the multi-step synthetic preparation of fluorescein derivatives requires starting materials with isomeric purity so that the reaction product can be easily purified and identified. |
