| Introduction | 7-chloro-1H-indole is a 7-halogenated indole. 7-halogenated indole is an important class of indole compounds. It has strong biological and pharmacological activities. Many natural or synthetic drugs and fine chemicals contain the structure of 7-halogenated indole, which has broad application prospects. |
| preparation | 3-chloro-2-nitrotoluene (2mmol) was dissolved in 20mL acetonitrile, DMFDMA(480mg,4 mmol) was added, and the temperature was raised and refluxed for 5h. TLC monitors the completion of the reaction, cooling, concentration under reduced pressure, and washing of the residue with water and a small amount of ethanol to obtain intermediate 4. Add intermediate 4 to a methanol solution 50mL 10% hydrazine, raise the temperature to 50 ℃, react for 8 hours, TLC monitor the reaction is completed, cool to room temperature, concentrate under reduced pressure, and separate and purify (petroleum ether: ethyl acetate 20:1) by column chromatography to obtain the target compound. The product detection data are as follows: white solid, 57% yield. |
