introduction | 7-chloroindazole is a pharmaceutical intermediate, which can be obtained from 3-chloro-2-hydroxybenzaldehyde as raw material and linked to hydrazine hydrochloride. it is reported in the literature that it can be used to prepare SYK inhibitors. |
Preparation | In a 250 mL flask, add hydrazine hydrochloride (excess) to a solution of 3-chloro-2-hydroxybenzaldehyde (36.7 mmol) in EtOH(30 mL). The solution was refluxed for 3 hours. The solvent is distilled under reduced pressure. The residue was purified by silica gel column chromatography (EtOAc∶PE = 10 ∶ 1) to give 7-chlorindazole. |
