In vitro study | Droxinostat was initially used as a sensitizer for FAS and TRAIL in PPC-1 cells by down-regulating the interleukin-1-like converting enzyme inhibitor protein (c-Fas), which is the region of death associated with c-FLIP. 20 μm -60 μm Droxinostat acts on PPC-1 cells in suspension culture, by first activating caspase 8 and then activating the mitochondrial pathway, which makes the cells more susceptible to apoptosis. Droxinostat also made other cancer cell lines including PC-3, DU-145, T47D, and OVCAR-3, but not LNCaP and MB-MDA-468, more susceptible to anodyne apoptosis, or CH-11-induced apoptosis. However, until now the direct target of Droxinostat has not been discovered. Droxinostat was found to selectively inhibit HDAC3, 6, and 8 in the histone acetylase (HDAC) 1-10 subtype, with IC50 values of 16.9 μm, 2.47 μm, and 1.46 μm, respectively, however, the effect on other HDAC members was not obvious, IC50 > 20 μm. 10 μM-100 μm Droxinostat acts on MCF-7 breast cancer cells to reduce cell survival and induce apoptosis by reducing the expression of c-FLIP L and c-FLIP S, making the cells more susceptible to apoptosis. |
