In vitro study | Importazole is a potent inhibitor of the Ran/importin-β interaction in vitro. Importazole specifically inhibited importin-β-mediated nuclear entry in Xenopus oocyte extracts and cultured cells, without disrupting transportin-mediated nuclear entry or CRM1-mediated nuclear exit. The addition of importazole during cell mitosis inhibits/disrupts the release of the importin-β cargo FRET probe, resulting in defects in spindle assembly. Importazole can inhibit cell proliferation and induce apoptosis of multiple myeloma cells by inhibiting NF-κB signaling pathway in vitro. |
