Method 1: Compound (I) and thionyl chloride are refluxed in methylene chloride to form chloride (II). Then it is condensed with potassium phthalamide in dimethylformamide, and the product (III) is reduced with hydrazine in ethanol to obtain nelacetam, which is neutralized with fumaric acid to obtain nelacetam fumarate.
Method 2: Compound (Ⅳ) is reacted with phosphorus oxychloride in hot dimethylformamide to dehydrate to obtain nitrile (V), and then hydrogenated and reduced under the catalysis of Raney nickel in ammonia-methanol mixture to obtain nelacetam, and neutralized with fumaric acid to obtain nelacetam fumarate.
